Abstract:Objective Toexplorethevalueofserum microRNA-372-3p (miR-372-3p)and microRNA-222-3p (miR-222-3p)levelsinpredictingadversepregnancyoutcomesinpatientswithpreeclampsia(PE).Methods Atotalof110 patientswithPEtreatedinNorthwestWomen'sandChildren'sHospitalfromFebruary2021toFebruary2024wereincluded asthePEgroup,andwerefurthergroupedintoamildPEgroup (n=41)andaseverePEgroup (n=69)basedontheir diseaseseverity.Meanwhile,108normalpregnantwomenwhounderwentprenatalcheckupswereselectedasthecontrol group.Thepregnancyoutcomeandrelatedclinicaldatawererecorded.Quantitativereal-timepolymerasechainreaction (qRT-PCR)wasusedtodetectserum miR-372-3pand miR-222-3plevels.Logisticregressionanalysiswasperformedto screenforinfluencingfactors.Receiveroperatorcharacteristic (ROC)curvewasplottedtoexplorethevalueofserum miR-372-3pandmiR-222-3plevelsinpredictingpregnancyoutcomesinPEpatients.Results TheserumlevelsofmiR-372-3p andmiR-222-3pinthePEgroupwerehigherthanthoseinthecontrolgroup(P<0.05).TheserumlevelsofmiR-372-3p andmiR-222-3pintheseverePEgroupwerehigherthanthoseinthemildPEgroup(P<0.05).Thelevelsof24hurinary protein,systolicbloodpressure,diastolicbloodpressureandserum miR-372-3pandmiR-222-3pinthepooroutcomegroup werehigherthanthoseinthegoodoutcomegroup(P<0.05).miR-372-3pandmiR-222-3pwereindependentriskfactorsfor adversepregnancyoutcomesinPEpatients.Theareaunderthecurve(AUC)ofthecombinedpredictionof miR-372-3pand miR-222-3p for adverse pregnancy outcomes in PE patients was higher than that of their single prediction (Z=2.804,2.186,P<0.05).Conclusion Theserum levelsof miR-372-3pand miR-222-3pin patients with PE are abnormallyelevated.Theelevatedlevelsofthetwoindicatorsareassociatedwithdiseaseprogressionandadversepregnancy outcomes.ThecombinationofthetwoindicatorscaneffectivelypredictadversepregnancyoutcomesinPEpatients.
